More Desmond

I am as excited as anyone to see what tricks Connor Kenway has up his sleeves, but I also want to spend a good amount of time with Desmond in the modern era. Assassin's Creed: Revelations ended with a twist that deserves ample time to resolve itself. Also, I desperately want to see Desmond scale some modern-day architecture.
More Customization

The customization features in previous games have grown to include quite a bit of variety, albeit mostly aesthetic. I would love to see an more options that shape Connor's and Desmond's fighting styles and even assassinations. With assassin recruits making some sort of return, it would also be great to have more control over these helpful characters and how they approach combat. Also, just because I am a sucker for visual choices, more clothing and armor customization would be great too.
Other Assassins

We know Assassin's Creed III: Liberation will feature series' first playable female assassin, but there are many interesting characters in the Order of Assassins that I would love to make short playable portions of Assassin's Creed III. The game is the culmination of long and epic struggle that has taken place over a millennium, it deserves the appearance of not just Connor Kenway, but a variety of assassin's that have played a part in the struggle against the Templars.
Warfare

The shadows are the natural home of Assassins, and stealth-kills their forte. But with American forces squaring off against the British, the revolutionary backdrop is too compelling to leave behind. Some trailers already show Connor Kenway rampaging through hostile forces with style all his own. If Ubisoft can seamlessly meld the large-scale battles with the traditional "assassin approach" to vanquishing enemies, then the revolutionary war will prove itself an excellent setting for Assassin's Creed III.
A Complete Conclusion

There is a huge amount of story to wrap-up with Desmond in this final game. We all know now what sort of reaction a less-than satisfactory conclusion means for fans of a series. I have faith Ubisoft can handle the conclusion of this story arc with tact, but it will be a thin road to walk with such a strong history and high expectations.
